摘要
目的观察川芎嗪注射液对血小板源性生长因子(PDGF)诱导的大鼠血管平滑肌细胞(VSMC)异常增殖过程中黏附分子信号传递的影响,以明确川芎嗪活血化瘀作用的新靶点。方法采用贴块法培养VSMC,建立PDGF诱导的VSMC增殖模型,用细胞计数法观察不同浓度川芎嗪对VSMC增殖的抑制作用。用Western印迹方法分别检测各组细胞间黏附分子(ICAM-1)及黏附分子信号传递蛋白——整合素耦联激酶(ILK)的表达变化及川芎嗪影响。结果与PDGF刺激组相比,川芎嗪呈剂量依赖性地抑制PDGF诱导的细胞增生,并显著降低ICAM-1〔仅相当于刺激组的44%,(P<0.05)〕及ILK〔仅相当于刺激组的62%,(P<0.05)〕的表达。结论川芎嗪通过抑制黏附信号传递及黏附分子的表达而发挥抗VSMC增生的作用。
Objective To investigate the Tetramethylpyrazine (TMP)-mediating anti-proliferation effect,and define its molecular mechanism in cultured rat aortic vascular smooth muscle cells (VSMC).Methods Cultured cells proliferating model of rats VSMC were established by platelet-derived growth factor (PDGF).Cultured cells were randomly divided into three groups:control,PDGF (20 ng/ml)and TMP treatment (20,40,80 μmol/L) groups.The number of VSMC were accounted under microscope.The expression of intercellular adhesion molecular-1 (ICAM-1) and integrin-linked kinase (ILK) were analyzed by Western blotting respectively.Results The proliferation of rat VSMC induced by PDGF could be inhibited by TMP significantly in a dose-dependent manner.A remarkable decrease of ICAM-1 and ILK expression were demonstrated in the TMP group compared with the PDGF group.Conclusions The anti-proliferation effect of TMP associates with suppression of adhesive molecules.
