血管内皮生长因子与地塞米松对早产鼠肺泡表面活性物质结合蛋白B表达的影响

Effect of Vascular Endothelial Growth Factor Combined with Dexamethasone on the Expression of Pulmonary Surfactant Associated Protein B of Premature Wistar Rats

摘要 目的:研究血管内皮生长因子(VEGF)以及地塞米松单独或联合应用对早产大鼠肺泡表面活性物质结合蛋白B(SP-B)mRNA表达的影响,及与肺发育的关系,为临床防治早产儿呼吸窘迫综合征(NRDS)提供新途径。方法:孕15d清洁级Wistar大鼠12只,随机分为4组,每组平均3只,试验分组:VEGF组、地塞米松组、VEGF加地塞米松组和对照组进行试验,应用RT-PCR检测早产鼠肺组织中SP-BmRNA的表达。结果:地塞米松组、VEGF加地塞米松组肺组织中SP-BmRNA表达与对照组比较,P<0.05,差异有显著性,VEGF加地塞米松组较地塞米松组SP-BmRNA表达进一步增加(P<0.05),而VEGF单独应用组与对照组比较,P>0.05,差异无显著性。结论:地塞米松能促进肺表面活性物质的合成和分泌,VEGF能协同地塞米松促进肺表面活性蛋白的分泌与合成。 Objective:To study the effect of vascular endothelial growth factor(VEGF)alone or combined with Dexamethasone on the development of lung of premature Wistar rats,and to explore a new way for preventing and treating the premature infant's respiratory distress syndrome.Methods:Wistar rats at 15 days of gestation were random divided into four groups:VEGF group,Dexamethason group,VEGF plus Dexamethason group,and control group.Lung tissue were harvested and extracted for total RNA,and mRNA levels of pulmonary surfactant associated protein B(SP-B)were determined by reverse transcription polymerase chain reaction(RT-PCR).Results:SP-B mRNA level in VEGF plus Dexamethason group was the highest,and that in Dexamethason alone group were also higher than the other two groups(all P0.05).No increase was found between VEGF group and control group in SP-B mRNA level in lung tissue.Conclusion:Dexamethason can increase pulmonary surfactant synthesis and secretion of the lung tissue,and VEGF can cooperate with Dexamethason to increase this effect.
