摘要
目的研究回复引导半胱氨酸丰富蛋白Kazal基元(reversioninducing cysteine rich protein with Kazal motifs,RECK)及基质金属蛋白酶(matrix metalloproteinases,MMP)2、9在宫颈癌中的表达,检测各种组织中的微血管密度(MVD),探讨三者之间的关系。方法采用免疫组化、细胞计数及测量灰度值的方法检测52例宫颈癌组织(实验组)及48例宫颈正常上皮、慢性宫颈炎组织(对照组)RECK、MMP-2、MMP-9基因的表达及MVD值。结果 RECK基因在宫颈癌组织的表达明显低于对照组(P<0.05);MMP-2、MMP-9基因在宫颈癌组织中的表达明显高于对照组(P<0.05)。实验组MVD值(19.4615±3.0718)明显高于对照组(12.0000±2.6629)(P<0.05);RECK的表达与MVD值呈负相关(r=-0.397,P=0.0495)。结论 RECK基因在宫颈癌组织中低表达,其可能抑制其微血管形成;MMP-2、MMP-9基因在宫颈癌组织的高表达,其可能促进其微血管形成,两者可能与宫颈癌转移有一定的关系。
Objective To detect the expression of RECK(reversion inducing cysteine rich protein with Kazal motifs),MMP-2 and MMP-9 in tissue of cervical carcinoma.MVD(microvascular density)were determined and the relationship of RECK,MMP-2,MMP-9 and MVD was measured.Methods Specimens were divided into two groups:52 cervical carcinoma cases were in Ca group and 48 patients with normal cervical tissue and benign cervical lesion consisted control group.S-P immunohistochemistry and integrated optical density(IOD)were used to detect the expression of RECK,MMP-2 and MMP-9.MVD were counted.Results The expression of RECK in Ca group significantly decreased and the Value of MVD in Ca group significantly increased compared to control group(P0.05).MVD negatively related to the expression of RECK(r=0.397,P=0.0495).In Ca group,the expression of MMP-2 and MMP-9 significantly increased(P0.05).Conclusions In Ca group,the expression of RECK markedly decreased while the value of MVD obviously increased and there were negative relationship between them.In cervical carcinoma,MMP-2 and MMP-9 are involved in the transition of cervical carcinoma and can promote angiogenesis of tumor.RECK,MMP-2 and MMP-9 are closely related to the prognosis of tumor.
