凋亡在寻常型天疱疮发病中的作用

Roles of apoptosis in the development of pemphigus vulgaris

摘要 以往认为寻常型天疱疮仅由致病性抗体作用导致的一类自身免疫性皮肤黏膜疾病.但近年来其发病机制不断被揭示,凋亡在疾病中的作用亦越来越明确,如角质形成细胞经抗体直接作用可产生凋亡现象;棘层松解后形成的细胞间连接丧失可触发失巢凋亡.松解凋亡的提出进一步阐明棘层松解和细胞凋亡之间的关联性,也证明寻常型天疱疮的凋亡与传统的凋亡现象不同,具有其特殊性,对寻找疾病治疗的新靶点具有重要意义. Pemphigus vulgaris was previously regarded as an pathogenic antibody-mediated autoimmune mucocutaneous disease. However, with the increase in knowledge about the pathogenesis of this entity, the roles of apoptosis in the development of pemphigus vulgaris have become more and more clear. For example, the apoptosis of keratinocytes can be induced by direct action of antibodies, and anoikis can be triggered by loss of cellular linkage following acantholysis. The hypothesis of apoptolysis further clarifies the correlation between apoptosis and acantholysis, and confirms that the apoptosis in pemphigus vulgaris is a unique paradigm of cell damage and detachment which should be distinguished from other classic known forms of cell death. These findings will facilitate the search of new targets for treatment of pemphigus vulgaris.
