肝动脉栓塞化疗与恩度联合应用对兔VX2肝移植瘤血管生成的影响 被引量:3

Effect of Transcatheter Arterial Chemoembolization Combined with Endostar on Angiogenesis of Transplanted VX2 Hepatic Tumor in Rabbits

摘要 目的探讨肝动脉栓塞化疗(Transcatheter arterial chemoembolization,TACE)与恩度联合应用对兔VX2肝移植瘤血管生成的影响。方法建立兔VX2肝移植瘤模型,并随机分为TACE组、抗血管生成组(TACE+动脉给予恩度)和生理盐水对照组。TACE术后14d,应用实时定量荧光PCR检测残癌组织血管内皮生长因子(Vascular endothelial growth factor,VEGF)mRNA的转录水平,免疫组化法检测肿瘤组织微血管密度(Microvessel density,MVD),并分析二者的相关性。结果TACE组VEGF mRNA的转录水平明显高于抗血管生成组和对照组(P均<0.05);抗血管生成组与对照组相比,差异无统计学意义(P>0.05)。抗血管生成组MVD(33.17±6.69)明显低于TACE组(59.96±12.19)和对照组(58.88±7.82),且差异均有统计学意义(P均<0.05);TACE组与对照组相比,差异无统计学意义(P>0.05)。各组肿瘤组织VEGF mRNA的转录水平与MVD的分布均呈正相关(r=0.40)。结论TACE与恩度联合应用与单纯应用TACE相比,可明显抑制肿瘤的血管生成。 Objective To investigate the effect of transcatheter arterial chemoembolization(TACE)combined with endostar on the angiogenesis of transplanted VX2 hepatic tumor in rabbits.Methods Rabbit model of transplanted VX2 hepatic tumor was established.The model rabbits were divided into three groups and treated by TACE,TACE combined with endostar and physiological saline(control)respectively.Fourteen days after TACE,the transcription level of vascular endothelial growth factor(VEGF)mRNA in residual tumor tissue was determined by real-time quantitative PCR,and microvessel density(MVD)of tumor tissue by immunohistochemical assay,based on which the relationship between the two results was analyzed.Results The transcription level of VEGF mRNA in tumor tissues of rabbits was significantly higher in TACE group than in TACE combined with endostar and physiological saline groups(both P 0.05),while showed no significant difference in TACE combined with endostar and physiological saline groups(P 0.05).However,the MVD of tumor tissue was significantly lower in TACE combined with endostar group(33.17 ± 6.69)than in TACE(59.96 ± 12.19)and physiological saline(58.88 ± 7.82)groups(both P 0.05),while no significant difference in TACE and physiological saline groups(P 0.05).The transcription level of VEGF mRNA in each group was positively related to the distribution of MVD(r = 0.40).Conclusion Compared with TACE alone,TACE combined with endostar inhibited the angiogenesis of tumor significantly.
