四妙勇安汤对实验性兔动脉粥样硬化易损斑块的影响 被引量:1

摘要 目的探讨四妙勇安汤对AS易损斑块的影响及机制。方法随机将40只日本大耳白兔分为正常组10只,实验组30只。正常组予普通饲料,实验组利用复合因素制备兔AS易损斑块模型,8周时实验组随机分为模型组、辛伐组、四妙组,24周取材。检测动脉壁内膜厚度(IT)、内中膜厚度比(IT/MT)、斑块纤维帽厚度(FCT)、纤维帽与内中膜厚度比(FCT/IMT)、胶原含量(CA)、α-平滑肌肌动蛋白(α-SMA)、基质金属蛋白酶-9(MMP-9)、肿瘤坏死因子-α(TNF-α)及核因子-κB(NF-κB)阳性面积百分比。结果模型组各指标与正常组比较,差异均有统计学意义(P<0.05,P<0.01)。四妙组IT、IT/MT均低于模型组(P<0.05),FCT和FCT/IMT均高于模型组(P<0.01);斑块内CA阳性面积百分比均高于模型组(P<0.01),α-SMA阳性面积百分比低于正常组MMP-9和NF-κB阳性面积百分比均低于模型组(P<0.01,P<0.05)。结论四妙勇安汤能够稳定AS易损斑块,其作用机制可能是通过抑制MMP-9、TNF-α及NF-κB表达实现的。 Objective To observe the intervening effect and acting mechanism of "Simiao Yongan Decoction" on extracellular matrix in rabbit' s atherosclerosis (AS) vulnerable plaque. Methods Forty Japanese white rabbits were randondy assigned into two groups: 10 rabbits in the normal group, and 30 rabbits in experiment group which was established to be AS vulnerable plaque model by high-fat diet combined with immune injury and femoral arterial balloon tearing. On the 8th week, 30 rabbits in experiment group were randomly divided into three groups: model group, "SimiaoYongan Decoction" group and simvastatin group, 10 rabbits in each group. On the 2gth week, the intimer thickness( IT), ratio of intimer thickness to media thickness( IT/MT), fiber cat thickness(FCT), FCT/IMT, collagen, α-SMA, tumor necrosis factor-α( TNF-α), matrix metalloproteinase-9 (MMP-9) and NF-κB expression in aortic wall were measured. Results Compared with the normal group, the IT, collagen, α-SMA and MMP-9 expression in aortic wall in the model group were higher (P 〈0.05) ; compared with the model group, in the "SimiaoYongan Decoction" group the IT, IT/MT, MMP-9 and NF-scB expression were lower(P 〈0.05, P 〈0.01 ), and FCT, FCT/MT, collagen and α-SMA were higher(P 〈0.05, P 〈0.01 ). Conclusion "SimiaoYongan Decoction" can stabilize atherosclerosis vulnerable plaque by inhibiting MMP-9, TNF-α and NF-κB expressions.
