摘要
目的:研究通痹合剂对佐剂型关节炎大鼠软骨细胞产生保护作用的机制。方法:培养正常大鼠软骨细胞,大鼠白介素(IL-1β)刺激后用ELISA法及阿利新蓝法检测10%含药血清对各组大鼠软骨细胞产生蛋白多糖(GAG)及Ⅱ型胶原的含量的影响。结果:通痹合剂组、消炎痛组、模型组佐剂型关节炎模型(AA)大鼠软骨细胞产生Ⅱ型胶原与正常组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组与正常组比较,差异无显著性意义(P>0.05)。通痹合剂组、消炎痛组产生Ⅱ型胶原与模型组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组、模型组及通痹合剂组与柳氮磺胺吡啶组分别比较,差异均无显著性意义(P>0.05);消炎痛组与柳氮磺胺吡啶组比较,差异有显著性意义(P<0.05)。通痹合剂组、消炎痛组、模型组AA大鼠软骨细胞产生GAG与正常组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组与正常组比较,差异无显著性意义(P>0.05)。通痹合剂组、消炎痛组产生GAG与模型组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组、模型组、通痹合剂组与柳氮磺胺吡啶组分别比较,差异均无显著性意义(P>0.05);消炎痛组与柳氮磺胺吡啶组比较,差异有显著性意义(P<0.05)。结论:通痹合剂对软骨细胞有保护作用,机制可能与其增加Ⅱ型胶原与GAG含量有关。
Objective:To explore the protective mechanism of Tongbi Mixture(TM) for chondrocytes of adjuvant arthritis(AA) rats.Methods:Normal rats chondrocytes were cultured and then were stimulated by interleukin 1β(IL-1β).After treatment with 10% drug-containing serum,the production contents of glycosaminoglycan(GAG) and type Ⅱ collagen in stimulated chondrocytes were detected by enzyme linked immunosorbent assay(ELISA) and alcian blue method.Results:Type Ⅱ collagen content in TM group,indomethacin group and model group differed from that in the normal group(P < 0.01),while the difference of type Ⅱ collagen content between sulfasalazine group and normal group was insignificant(P > 0.05).Type Ⅱcollagen content in TM group and indomethacin group differed from that in the model group(P < 0.01),while the difference of type Ⅱcollagen content in sulfasalazine group with model group and TM group with sulfasalazine group was insignificant(P > 0.05).Type Ⅱ collagen content in sulfasalazine group differed from that in indomethacin group(P < 0.05).GAG in TM group,indomethacin group and model group differed from that in the normal group(P < 0.01),while the difference of GAG between sulfasalazine group and normal group was insignificant(P > 0.05).GAG in TM group and indomethacin group differed from that in the model group(P < 0.01),while the difference of GAG in sulfasalazine group with model group and TM group with sulfasalazine group was insignificant(P > 0.05).GAG in sulfasalazine group differed from that in indomethacin group(P < 0.05).Conclusion:TM exerts protective effect on chondrocytes of adjuvant arthritis rats,and its mechanism may be related with the increase of type Ⅱ collagen and GAG contents.
