针刺对胰岛素抵抗模型大鼠骨骼肌PI3K p85的影响 被引量:10

Effect of Acupuncture on PI3K p85 Expression in Skeletal Muscle of Insulin-resistance Rats

摘要 目的:观察针刺对胰岛素抵抗模型大鼠骨骼肌磷脂酰肌醇3激酶(PI3K)之亚基p85的影响。方法:将24只SD大鼠随机分为空白组、模型组和针刺组各8只,以高糖高脂高盐饮食复制胰岛素抵抗动物模型,采用葡萄糖氧化酶法、ELISA、实时荧光定量PCR、western-blotting等方法,检测比较各组大鼠空腹血糖(FBG)、血浆胰岛素(FINS)、胰岛素敏感性指数(ISI)、血清C-肽(C-P)和骨骼肌PI3K p85α mRNA及蛋白的表达。结果:模型组大鼠的FBG、FINS、C-P、PI3K p85α mRNA、PI3Kp85蛋白表达均较空白组显著升高(P<0.01),ISI显著降低(P<0.01);针刺组的FBG、FINS、C-P、PI3K p85α mRNA、PI3K p85蛋白表达均较模型组有不同程度的降低(P<0.05,P<0.01),ISI显著升高(P<0.01)。结论:针刺干预可以纠正胰岛素抵抗模型大鼠骨骼肌PI3K p85蛋白的过度表达,使PI3K的活性升高,这可能是针刺改善胰岛素抵抗的作用机理之一。 Objective:To observe the effect of acupuncture on subunit p85 of phosphatidyl inositol kinase 3(PI3K) in skeletal muscle of insulin-resistance rats.Methods:Twenty-four SD rats were randomized into blank group,model group and acupuncture group.Rat models of insulin resistance were induced by feeding of high glucose,high fat and high salt diet.Glucose oxidase method,enzyme linked immunosorbent assay(ELISA),real-time fluorescent quantitative polymerase chain reaction(PCR),and western-blotting method were used for the detection of fasting plasma glucose(FPG) content,fasting plasma insulin(FINS) level,insulin sensitivity index,serum C-peptide(C-P) and PI3K p85 mRNA and protein expression in skeletal muscle of different groups respectively.Results:Compared with the blank group,FPG,FINS,C-P,PI3K p85α mRNA expression and PI3K p85 protein expression were increased,while ISI was decreased in the model group(P < 0.01).FPG,FINS,C-P,PI3K p85α mRNA expression and PI3K p85 protein expression were decreased(P < 0.05 or P < 0.01),while ISI was increased in acupuncture group(P < 0.01).Conclusion:Intervention of acupuncture can regulate the over-expression of PI3K p85 protein in skeletal muscle of insulin-resistance rats,enhance the activity of PI3K,which may be one of the therapeutic mechanisms of acupuncture for insulin resistance.
