摘要
目的:探讨大蒜素诱导卵巢癌细胞株OVCA-3凋亡规律。方法:应用DNA断裂点标记(TUNEL)法、DNA凝胶电泳以及光镜等技术观察有关细胞的凋亡情况。结果:0.01~100μg/ml的大蒜素作用OVCA-3细胞2h及10μg/ml的大蒜素作用OVCA-3细胞不同时间,锥虫蓝染色阳性率均在18.5%以下。TUNEL显示:1~100μg/ml的大蒜素作用OVCA-3细胞2h,即出现典型的细胞凋亡特征;50μg/ml时凋亡率为65.9%。结论:一定剂量范围内的大蒜素可在直接杀伤作用较小的情况下明显诱导OVCA-3细胞凋亡,不会因细胞坏死而引起炎症反应,这对于大蒜素应用于临床将有重要意义。
Objective: To investigate the rule of allicin induced apoptosis. Methods: Terminal
deoxylnucleotide transferase directed XUTP nick and end labelling (TUNEL) assay, DNA gel
electrophoresis and light microscopy were used to observe apoptosis. Results: Trypan Blue
staining positive rates of OVCA3 cell were under 18.5% after being exposed to allicin at the
dosage 0.01 ̄100 g/ml for 2 h and 10 g/ml for different time. Allicin induced apoptosis of OVCA3
cells at 1100 g/ml for 2 h. Apoptosis peak was shown at 50 g/ml. Apoptotic cells accounted for
65.9% of the population. Z]Conclusion: Allicin of appropriate dosage induces apoptosis of
OVCA3 cell, while has little toxicity on OVCA3 cell. That is important for clinical application of
allicin.
