摘要 本文优化了反相高效液相色谱测定葡萄酒有机酸的色谱条件。研究表明,采用Waters Atlantis T3反相C18色谱柱(4.6mm×150mm,5μm),保护柱为Waters Atlantis T3 Security Guard(4.6mm×1.5mm,5μm),含1%甲醇的0.02mol/L磷酸二氢钾(pH2.7)为流动相,等度洗脱,流速为0.8mL/min,UV紫外检测器,柱温40℃,检测波长为210nm和243nm,能将7种有机酸有效分离。精密度和回收率分析表明,该方法操作简单、准确率高。 The chromatographic conditions for the analysis of wine organic acids using reversed phase high performance liquid chromatography (RT-HPLC) was optimized in this study. The result showed that seven organic acids in wine could be effectively separated and detected under the following conditions: Waters Atlantis T3 RP-CI8 colume (4.6mm × 150mm, 5 μm), Waters Atlantis T3 Security Guard (4.6ram ×1.5mm, 5 μm), a mobile phase of 0.02mol/L KH2PO4 (pH2.7), isocratic elution with a flow rate of 0.8 mL/min, a column temperature of 40℃ and detection wavelengths at 210nm and 243nm. This method was demonstrated to be simple in operation, accurate in detection and have a high collection of sample.
