肿瘤浸润淋巴细胞与IL-2联合诱导Lewis肺癌凋亡的研究

Study on anti tumor effect of tumor infiltrating lymphocytes from lung cancer combining with IL 2: induce apoptosis in vivo

目的：研究肿瘤浸润淋巴细胞（ＴＩＬ）与ＩＬ２联合体内诱导凋亡产生抗瘤作用机理，为应用ＴＩＬ治疗肺癌提供依据。方法：从Ｌｅｗｉｓ肺癌（ＬＬＣ）瘤体中分离提取ＴＩＬ，在含１０００Ｕ／ｍｌＩＬ２的完全培养基中培养后，与ＬＬＣ细胞以１０∶１接种于Ｃ５７ＢＬ／６小鼠腋下，每日１０００Ｕ／ｍｌＩＬ２瘤内注射，并测量肿瘤体积，于接种后第１８天处死小鼠。结果：发现ＴＩＬ加ＩＬ２有明显地抑制ＬＬＣ生长的作用（Ｐ＜０００１），ＨＥ染色、ＴＵＮＥＬ和电镜均证实ＴＩＬ加ＩＬ２可明显诱导Ｌｅｗｉｓ肺癌细胞凋亡。

Purpose To study the mechanism of anti tumor in vivo of tumor infiltrating lymphocytes(TILs). Methods TILs isolated from Lewis lung cancer (LLC) implanted in C57BL/6 mice, which were cultured with IL 2, were implanted into C57BL/6 mice together with LLC cells, which the ratio of TIL: LLC cells was 10∶1. These mice were injected 1000 U IL 2 and the tumor size were measured. On the eighteenth day after the cells having been implanted, the mice were killed. Results The data suggested that TILs plus IL 2 can significantly suppress tumor growth ( P <0 01). Histologically, TUNEL and ultrastructure confirmed that TILs combining with IL 2 had the effect of inducing the presence of LLC apoptosis. Conclusion The results indicate that inducing apoptosis in vivo may be one of the mechanism of the anti tumor effect of TILs combining with IL 2.
