中药复方AK粗提物协同顺铂抗结肠癌动物模型研究

Experimental study on anti-colorectal cancer efficacy of extracts of TCM Compound AK combined with cisplatin

目的:探讨中药复方AK粗提物对化疗药物顺铂的减毒增效作用。方法:建立BALB/c小鼠CT-26结肠腺癌动物模型,随机分为6组,A组为对照组,B组为单用顺铂组,C组为单用AK组,D、E和F组为协同实验组。建模24h后进行干预,连续用药12d。第14d处死小鼠,观察抑瘤率、血常规、胸腺指数、脾脏指数、肝肾功能,流式细胞术检测外周血T细胞亚群,显微镜下观察移植瘤和肾组织的变化。结果:①与对照组比较,D、E、F各实验组瘤重明显降低(P<0.05);B、C、D、E、F各组抑瘤率分别为55.92%、28.11%、61.05%、71.11%、69.72%,协同实验组抑瘤率明显增高。②与对照组比较,B组的白细胞、血小板数量降低,BUN升高(P<0.01);与B组比较,D、E、F组白细胞、血小板数量升高,BUN降低(P<0.01)。③与对照组比较,B组CD3+、CD4+、CD4+/CD8+比值明显降低,CD8+值升高(P<0.05);与B组比较,D、E、F组CD3+、CD4+、CD4+/CD8+值有所提高,CD8+值降低(P<0.01)。④与对照组比较,B组小鼠胸腺指数和脾脏指数明显降低(P<0.05);D、E、F组的小鼠胸腺指数和脾脏指数均高于B组(P<0.05)。结论:中药复方AK粗提物对顺铂具有减毒增效的作用。

Objective：To investigate the toxicity-reducing and synergistic effects of the extracts of AK prescription for chemotherapy.Methods：Models of colorectal CT-26 tumor bearing mice were established and randomly divided into six groups,including group A（control group）,group B（CDDP group）,group C（AK group） and combined treatment groups D,E and F.The medication was begun after 24 hours since inoculation,once a day for 12 days.Mice were sacrificed on day 14th.The tumor inhibitory rate,thymus index,spleen index,liver and renal function and the blood cell in the peripheral blood were measured.The subtypes of T lymphocytes in the peripheral blood were detected by flow cytometry.Optical microscopy was employed to observe the structure of tumor and renal.Results：①The tumor weight in groups D,E and F was lighter than that in group A（P0.05）.Groups B,C,D,E and F could inhibit the tumor growth and the tumor inhibition rate was 55.92%,28.11%,61.05%,71.11%,69.72%,respectively.②Compared with group A,in group B the count of WBC and PLT was decreased obviously and the level of blood urea nitrogen（BUN） was increased（P0.01）.Compared with group B,in groups D,E and F the count of WBC and PLT was increased and the level of BUN was decreased（P0.01）.③Compared with group A,in group B the level of CD3＋,CD4＋,CD4＋/CD＋8 was lower and the level of CD＋8 was higher（P0.05）.The level of CD3＋,CD4＋,CD4＋/CD＋8 in groups D,E and F,was higher than that in group B,but CD＋8 value was decreased（P0.01）.④Compared with group A,thymus index and spleen index were decreased in group B（P0.05）.Groups D,E and F had different levels ofincrease in thymus index and spleen index compared with group B（P0.05）.Conclusion：The extracts of prescription of TCM AK can display the toxicity-reducing and synergistic effects for chemotherapy.
