摘要
采用cDNA-AFLP技术分析了4℃低温诱导前后耐寒辣椒(Capsicumannuum L.)品系P70的基因表达差异谱。结果获得了120个差异片段,对阳性克隆中的12个差异片段进行测序和Blast-x比对,发现其中有4个片段(KH-1、KH-2、KH-3和KH-4)与抗逆性相关,其碱基数分别为185、160、269和511 bp,4个片段分别与编码抗坏血酸过氧化物酶基因的同源性达98%、与细胞色素p450基因的同源性达98%、与牛血清蛋白(BSA)基因的同源性达94%、与水孔通道蛋白(AQP)基因的同源性达90%。4个差异基因表达模式为:基因KH-1、KH-2、KH-3上调表达,KH-4下调表达。
P70(Capsicum annuum L.) is a pepper line with excellent chilling-tolerance.The gene expressions of P70 stressed at 4 ℃ for different times were studied via cDNA-AFLP.One hundred and twenty differential expression cDNA fragments were obtained,twelve positive fragments of which were sequenced and analyzed by Blast-x.The blast showed that four sequenced fragments(KH-1 about 185 bp,KH-2 about 160 bp,KH-3 about 269 bp and KH-4 about 511 bp) were similar to the known genes of chilling-tolerance in the database,in which KH-1,KH-2,KH-3 and KH-4 had high homology to ascorbic acid peroxidase gene(98%),cytochrome p450 gene(98%),bovine serum albumin(BSA)gene(94%)and water-hole channel protein(AQP) gene(90%),respectively.The differential expression of cDNA also showed that KH-1,KH-2,KH-3 were up-regulated and KH-4 was down-regulated under the cold stress.These genes will reveal the mechanism for cold tolerance.
