胡卢巴等位酶变异和遗传多样性研究 被引量:5

摘要 采用聚丙烯酰胺凝胶电泳技术,对33份胡卢巴(Trigonella foenum-graecum L.)种质资源的5个酶系统24个酶位点的变异和遗传多样性进行初步分析。结果表明:参试胡卢巴的遗传多样性水平较高,物种水平上的遗传多样性参数:多态位点百分数(P)为75.0%,平均每位点的等位基因数(A)为1.75,平均期望杂合度(He)为0.352;种源水平上的平均遗传多样性参数:P=70.6%,A=1.87,He=0.422。各种质资源间基因分化系数(GST)为0.026;基因流(Nm)的平均值为26.14,表明胡卢巴不同种质资源间基因分化水平低,基因交流频繁。聚类分析结果显示,来自中国不同地区的胡卢巴亲缘关系较近,而国外胡卢巴和国内胡卢巴的亲缘关系相对较远。 Allozymic variation and genetic diversity of Fenugreek(Trigonella foenum-graecum L.) were evaluated for 33 germplasm resources using vertical slab polyacrylamide gel electrophoresis.A total of 24 loci from 5 enzyme systems(Esterase,Est;Malic enzyme,Me;Malate dehydrogenase,Mdh;Peroxidase,Per;Shikimate dehydrogenase,Skd) were detected,and a high level of genetic diversity was revealed both at the species level(percentage of polymorphic loci P=75.0%,mean number of alleles per locus A=1.75,mean expected heterozygosity He=0.352) and at population level(P=70.6%,A=1.87,He=0.422).The coefficient of gene differentiation(GST) among 33 germplasm resources was 0.026,and the mean gene flow(Nm) was 26.14,which indicated that there were frequently genes exchange and low level of genetic differentiation among different germplasm resources.Cluster analysis showed that there was a close relative relationship in fenugreek which from different regions in China,whereas the relationship between fenugreek germplasm resources from abroad and domestic was relatively far away.
