摘要
探讨了用油桐尺蠖成虫卵巢细胞系 (Bs4 84细胞系 )增殖油桐尺蠖核型多角体病毒 (BsNPV)的若干工艺条件。结果显示 ,病虫血淋巴在不同温度条件下保存 ,4℃一个月内 ,- 2 0℃和液氮中 4~ 6个月内 ,病毒仍具有实用水平的活性 ;感染后的细胞培养物 ,用超声波破碎优于液氮冻融法 ;感染后的细胞培养物培养2 4 ,36 ,4 8,6 0 ,72h取样 ,超声波破碎 ,培养 6 0h的样品具有较高病毒滴度 ;于感染后第 4 ,5,6 ,7d取样 ,细胞不破碎 ,第 5~ 6天病毒滴度达较高值。
This paper reports some concrete methods for using BS-484 cell line to reproduce buzura suppressaria larvee infected by BsNPv was Preserved for 4~6 months,at -20 ℃,or in liquied nitrogen.To release cell—associated nuclocapsids ,the infected cell culture were lysed by sonication or by freaza-thawing.Sonication is the more excellent method.The infectivity of cell culture lysed by sonication reachs 10 5.68 TCID 50 ·ml -1 when infected for 60 hours ,while the infectivity of cell culture no lysed reachs 10 6.17 TCID 50 ·ml -1 when infected for 6 days.
