摘要
以杏嫩叶为材料,利用改进的CTAB法,提取到高质量的杏叶片总DNA,通过优化酶切、连接、预扩增、选择性扩增等试验条件,建立了高效稳定的杏荧光AFLP反应体系;筛选出多态性好的12对引物组合,均获得稳定、清晰的扩增图谱,可进行杏品种鉴定、种质资源的遗传多样性和亲缘关系分析等研究。
The high quality DNA required for construction of AFLP analysis system was isolated with an improved method of CTAB from young leaves of apricot.A fluorescent-AFLP reaction system was constructed for genetic analysis of apricot by optimizing DNA restriction-ligase reaction,pre-amplification and selective amplification.The clear and stable amplification patterns could be obtained from 12 pairs primer combinations with good polymorphism,which were selected from 64 pairs AFLP primer combinations.These results might provide a method based on the fluorescent-AFLP system for the further study in the field of molecular marker in cultivar identification,genetic diversity and genetic relationship in apricot.
