摘要
目的:探讨p38反义寡核苷酸(p38 anti-ODN)对卵巢癌细胞株(CAOV3)增殖的影响。方法:培养卵巢癌细胞株(CAOV3),以MTT比色法及克隆形成实验测定p38 anti-ODN抑制卵巢癌细胞株CAOV3增殖的作用。免疫印迹法测定p-p38、Cyclin D1及p21waf/cip1表达。免疫沉淀法纯化蛋白并p38试剂盒测定p38活性。结果:MTT实验及克隆分析法均显示p38 anti-ODN明显抑制CAOV3细胞增殖、p-p38表达、下调Cyclin D1表达,同时上调p21waf/cip1表达。结论:p38反义寡核苷酸可以抑制卵巢癌细胞株(CAOV3)增殖,其机制与影响p38通路及下游CyclinD1、p21waf/cip1表达有关;干预p38通路是基因治疗卵巢癌的重要途径之一。
Objective:To observe the effects of p38 anti-ODN on the proliferation and p38 signal transduction of human ovarian cancer CAOV3 cell line.Methods:The effect of p38 anti-ODN on the proliferation of CAOV3 cells was observed by MTT assay and clone formation experiment. The expressions of p38, cyclin D1, p21^waf/cip1 were analyzed by Western blot. p38 activity was measured by immuno-precipitation.Results:p38 anti-ODN could depress the proliferation of CAOV3 cell line. The intracellular and p38 expression were decreased, the expression of Cyclin D1 were decreased, while p21^waf/cip1 expression was elevated by different concentrations of p38 anti-ODN.Conclusion:p38 anti-ODN can depress the proliferation of CAOV3 cell line with inhibiting p38 signal transduction; targeting to p38 signal transduction is a useful method to curing ovarian cancer.
