摘要
用SSP-PCR的方法在视网膜库中进行扩增,结果在RPGRcDNA的5端发现了一段60bp的新顺序,该序列可以延伸原有的读框。通过在网上数据库中的寻找和比较,在RPGR基因的15号内含子内发现了一段长588bp的表达顺序,进一步的实验表明该序列在视网膜中也表达。研究结果提示有另外的RPGR转录产物存在,该研究为寻找RPGR基因的另外转录产物奠定了基础。
By using SSP (single specific primer) PCR in retinal library, a novel segment of RPGR cDNA sequence in the 5 end, which can extend the ORF, was found. A 588bp segment expressed sequence in the intron 15 of RPGR gene was also obtained by searching database. This sequence is also expressed in retinal library.The results support the hypothesis that maybe there is other spliced product of RPGR gene in retinal tissue. This work laid a foundation in finding a new transcribed product of RPGR gene.
