以红瑞木茎段为外植体,研究了其组织培养技术。结果表明:适合红瑞木芽启动培养的培养基为MS+BA0.5 mg·L^(-1)+IBA0.5 mg·L^(-1),诱导率可达93%;增殖培养基以MS+BA0.5mg·L^(-1)+IBA0.3 mg·L^(-1)为最佳,增殖系数可达4.15;生根诱导的最适培养基为1/4MS+IBA0.05 mg·L^(-1),生根率可达90%;生根苗在温室条件下成活率可达80%以上。
An efficient method for in vitro regeneration of Comus alba using stem-segment as explants via adjusting the ratio of IBA-BA presented in the MS medium was described. The results showed that MS medium supplemented with 0. 5 mg · L-1BA and 0. 5 mg · L-11BA was the most effective treatment in shoot initiation while 0.5 mg · L-IBA and 0.3 mg · L-1IBA in shoot multiplication, providing the highest shoot induction frequencies (93 %) and the highest coefficient of shoot multiplication (4.15), respectively. The highest rooting rate was achieved using 1/4 MS medium in combination with 0.05 mg· L-11BA. Over 80% of the regenerated plantlets successfully acclimatized to greenhouse conditions. This system provides a solid basis for large-scale multiplication of C. alba by tissue culture.
