幽门螺杆菌与胃癌BGC-823细胞凋亡

EFFECTS OF HALICOBACTER PYLORI ON APOPTOSIS OF HUMAN GASTRIC CANCER CELL LINE BGC-823 IN VITRO

摘要 目的研究幽门螺杆菌(Halicobacter pylori,Hp)在胃癌细胞BGC-823凋亡中的作用及作用机制。方法将BGC-823细胞与Hp以不同浓度培养,倒置显微镜观察细胞形态学变化;透射电镜观察细胞凋亡及超微结构变化;流式细胞仪检测细胞凋亡率。结果与对照组相比,低中浓度Hp作用BGC-823细胞后,倒置显微镜观察到细胞增殖旺盛,并出现明显的凋亡抑制现象;电镜观察到细胞凋亡形态特征及凋亡数目减少。流式细胞仪检测结果:与对照组相比,中低浓度组凋亡率下降,有显著性差异(P<0.05),高浓度组细胞的凋亡率上升,有显著性差异(P<0.05),以上作用均呈细菌浓度和作用时间依赖性。结论 Hp可抑制胃癌细胞BGC-823凋亡,这可能与凋亡抑制基因上调有关。 Objective To investigate the effect of Halicobacter pylori(HP) on apoptosis of gastric cancer cell line BGC-823.Methods The BGC-823 cells were co-cultured in different concentration under Halicobacter pylori presenting.Flow cytometric analyses were used for the assessment of apoptosis rates after infection.Cell ultra-structure changes and apoptosis feature were observed by the transmission electron microscope and light microscope.Results Compared with controls,proliferation of cells in the low and middle concentration of Halicobacter pylori groups shown well and the apoptosis was inhibited.pre-apoptosis feature were appeared under transmission electron microscope.assessment of apoptosis rates detected by flow cytometry showed that cell apoptosis rate of the low and middle concentration of Halicobacter pylori groups had a tendency of decreased rates(P﹤0.05).However,in high concentration group,cell apoptosis rates were remarkableincreased(P﹤0.05),apoptosis depends on the time of HP infection and the concentration HP.Conclusion Infection of H.pylori can inhibit apoptosis of gastric cancer cell BGC-823,the mechanisms may be related to up-regulation of apoptosis inhibition gene.
