CD4^+单核细胞中HIV-1 gp120与gp160质粒的构建与表达

The Expression of HIV 1 gp120 and gp160 in Human CD4 + Monocytoid Cells

采用ｐＳＭＴＥ７作为质粒，进行ＨＩＶ－１ｅｎｖ基因对ＣＤ４阳性单核细胞Ｕ９３７－２的导入与表达，ｅｎｖ基因导入细胞在微量重金属镉的诱导下，激活启动子表达ｇｐ１２０及ｇｐ１６０，经Ｗｅｓｔｅｒｎｂｌｏｔ转移显色后，可得到清晰的ｅｎｖ目的条带．此外，通过流式细胞仪对表达的ｇｐ１２０与ＣＤ４位点的结合作了分析，验证了该表达蛋白的结构与活性．从实验系可以观察到表达ｇｐ１６０的ＣＤ４细胞株由于表达ｇｐ１６０而引起细胞死亡，而表达ｇｐ１２０的细胞株在表达ｇｐ１２０时不受任何影响．

Studies were made on the expression of HIV 1 gp120 and gp160 in human CD4 + monocytes. The cloned cell lines were induced with metal ions and expressed the gp120 and gp160 of HIV 1. The structure and activity of these proteins were proved by SDS PAGE, Western blot and immunoprecipitation analyses. Cell death was found in CD4 +cell while expressing gp160 but no apparent cytopathic effect was found in CD4 + cell while expressing gp120.
