摘要
目的本文考察了壳聚糖絮凝法用于澄清五苓滴丸的最佳工艺。方法对絮凝条件进行了优化,选择干浸膏收率、总多糖含量两个指标与醇沉工艺进行了比较,其中总多糖含量的测定采用蒽酮-硫酸比色法。结果在有效成分的保留率上,壳聚糖絮凝法明显优于醇沉法。优选的最佳澄清工艺为药液与药材的浓缩比例为4∶1,壳聚糖用量为0.8%,于40℃下,以50 r/min的速度,搅拌10 min。结论壳聚糖絮凝法不仅澄清效果好,而且经济实用,可代替醇沉法用于五苓滴丸的澄清。
Objective To study the optimum clarification process of Wuling dripping pills by chitosan.Methods The operating conditions were optimized,and the results were compared with alcohol precipitation method according to the dry-extracts content and the content of total polysaccharides.The content of total polysaccharides was measured by anthrone-sulfuric acid colorimetry.Results The retaining of active compounds of chitosan flocculating method was better than that of alcohol precipitation method.The optimized flocculation technique conditions:the concentration ratio of decoction was 1∶ 4,the amount of adding chitosan was 0.8%,and flocculation temperature was 50 ℃,mixing 10 min with the rate of 50 r/min.Conclusion Chitosan flocculating method was efficient and economic on the clarification of Wuling dripping pills,and can replace the alcohol precipitation method.
