NMDA受体拮抗剂APV抑制福尔马林诱导的大鼠伤害性行为反应

摘要 目的:检测NMDA受体拮抗剂DL2amino5phosphonovalericacid(APV)对福尔马林诱导的大鼠伤害性行为反应的镇痛效应。方法:在大鼠左足趾部sc福尔马林诱导化学损伤性行为反应,经蛛网膜下腔引入药物,观察对此行为反应的抑制效应。结果:APV剂量依赖地抑制福尔马林诱导的持续性伤害性行为反应并有时间作用特点;APV对其中的反射性缩腿反应也有抑制作用,但没有时间作用特点。结论:NMDA受体在脊髓痛觉传递和疼痛反应过程中起重要作用;但NMDA受体拮抗剂APV是否有直接的镇痛效应仍需进一步分析。 AIM: To check the analgesic effect of DL2amino5phosphonovaleric acid (APV), an NMDA receptor antagonist, on the nociceptive behavioral responses provoked by formalin. METHODS: Inject subcutaneously 50 l of 25% formalin into the plantar of the rat hindfoot. Administer APV intrathecally and then analyze possible inhibitory effect of APV on nociceptive behavioral responses. RESULTS: APV dosedependently inhibited these persevering nociceptive behavioral response, the effect was characterized by a timereaction; but the inhibition of flinching response led by APV showed no characteristic for timereaction. CONCLUSION: NMDA receptor importantly participates in the neural transmission and modulation of nociception in the spinal cord. Whether or not NMDA receptor antagonist (APV) play direct role in ameliorating nociceptive behavioral responses should be further ascertained.
