不同体外微环境条件对精子生存质量的影响 被引量:1

摘要 目的探索精子生存质量最佳的体外微环境条件。方法收集禁欲5~7 d的精液标本30份,精液处理前将梯度离心液和冲洗液分别放在37℃恒温水浴箱(甲组)和室温(乙组)中复温,室温下液化后以密度梯度离心处理并且洗涤1次,然后分别放在室温(条件A)、37℃6%CO2培养箱(条件B)、37℃恒温水浴箱(条件C)3种不同培养条件下保存,于24 h、48 h和72 h检查精子的存活率。结果培养液在甲组中复温的精子24 h、48 h和72 h的存活率(74.0±9.4)%、(68.2±13.6)%(、52.9±26.5)%,略高于乙组的(70.5±16.2)%、(61.1%±15.8)%、(50.5%±17.4)%,但两组比较差异无统计学意义(P〉0.05)。培养液在甲组或乙组复温并处理后,精子在体外微环境条件下培养随着时间延长其存活率均有降低趋势,而在保存时间相同情况下,培养在室温(条件A)中的精子与培养在37℃恒温水浴箱(条件C)内的精子存活率比较差异无统计学意义(P〉0.05),但是培养在37℃6%CO2培养箱(条件B)内的精子存活率显著低于培养在室温或37℃恒温水浴箱内的精子的(P〈0.05)。结论精液处理前所用培养基最好放置于37℃恒温水浴箱中复温,处理好的精子应立即做授精准备。 Objective In order to explore the best micro-environmental conditions for sperms viability quality in vitro.Methods 30 semen samples were collected from 30 men whose abstinence period was between 5-7 days,then gradient centrifugation liquid and washing liquor were gradiently centrifugate and washed by the sperm washing medium which were rewarmed in 37℃ water incubator(group one)and at room temperature(group two)after liquefaction at the room temperature.Sperm viability was checked at 24 hours,48 hours,and 72 hours respectively after cultured in three different micro-environmental conditions at room temperature(condition A),37℃ 6% CO2 incubator(condition B),37℃ water incubator(condition C).Results The survival rate of the sperm whose medium was rewarmed in group one were(74.0±9.4)%,(68.2±13.6)%,(52.9±26.5)% slightly higher than those of group two(0.5±16.2)%,(61.1%±15.8)%,(50.5%±17.4)%,but the differences were not statistically significant(P0.05).The survival rate of sperm which cultured in different micro-environmental conditions in vitro after rewarming declined linear by time.In the same culture time,the survival rate of sperm cultured at room temperature(condition A)was no significant differentce with which cultured in water bath(condition C)(P0.05),but the sperm viability cultured at 37℃ 6% CO2 incubator(condition B)was significantly lower than that at 37℃ water bath or at room temperature(P0.05).Conclusion All culture medium which would be used to treat the semen before treatment is best placed with medium temperature water bath at 37℃ in the rewarming.The sperm should be used to fertilize immediately after being treated.
