摘要
目的观察胰高血糖素样肽-2(GLP-2)对小鼠小肠黏膜的作用效果;检测胰高血糖素样肽-2受体(GLP-2R)在小鼠不同脏器的分布和表达。方法选用5-6周龄的雄性健康BALB/C小鼠,体质量17-20 g。随机分为3组,腹腔注射组9只,脑内注射组9只,对照组8只,分别给予相应处理,3 d后处死小鼠,做组织切片进行HE染色,观察小鼠小肠黏膜的组织学变化,用免疫组化方法检测GLP-2R在不同脏器的表达和分布。结果①小鼠经腹腔注射GLP-2后,小肠不同肠段黏膜的绒毛高度较对照组明显增加(P均〈0.05);脑内注射组的肠黏膜无明显形态学变化。②GLP-2R在小鼠胃、空肠、结肠、气管等部位均有表达,而食管、肝、肺、肾与膀胱显阴性。③腹腔注射组的GLP-2R表达较对照组显著下调(P〈0.05),而脑内注射组的GLP-2R表达无变化。结论 GLP-2能刺激小肠黏膜上皮增厚,增加小肠不同肠段黏膜的绒毛高度;小鼠的胃、空肠、结肠、气管均有GLP-2R的表达,食管、肝、肺、肾与膀胱无GLP-2R的表达。
Objective It is to observe the effect of GLP-2 on mucous membrane of small intestine and detect the expression and distribution of GLP-2R in the different parts of different organs in mice. Methods Male and healthy BALB/C mice(5 to 6 weeks old) weighing 17 to 20g were used and divided into control group(n=8),GLP-2 cerebral cavity treatment group(n=9) and GLP-2 cavum abdominis treatment group(n=9) randomly.All animals were treated with corresponding method.Three days later,the mice were sacrificed.The histological changes of mucous membrane of small intestine in mice were observed by dyeing with hematoxylin and eosin staining,and the expression and distribution of GLP-2R in gastrointestinal tissue were detected by Immunohistochemistry.Results ①After cavum abdominis treating with GLP-2,the mucosal villus in different section of small intestine grow obviously higher compared with control group(P〈0.05);and no obvious morphological changes was found in cerebral cavity treatment group.②GLP-2R was widely expressed in each part of gastrointestinal tract,trachea in mice.But it was not observed in esophagus,liver,lung,kidney and bladder.③GLP-2R in cavum abdominis treatment group was less obvious compared with the control group(P〈0.05) while GLP-2R in cerebral cavity treatment group has no changes.Conclusion GLP-2 is able to stimulate small intestine epithelium mucosa to grow and increase mucosal villus height in different section of small intestine;GLP-2R is widely expressed in gastric,jejunum,colon,and trachea in mice,the expression became less obvious;the expression of GL P-2R in esophagus,liver,lung,kidney and bladder was not observed.
