摘要
目的观察脾虚证大鼠唾液淀粉酶分泌情况及四君子汤治疗作用。方法雄性SD大鼠随机分为正常组、模型组,模型组,大鼠皮下注射利血平,空白组大鼠注射等量生理盐水,共8d;造模结束后将模型组分为中药组和模型组,中药组给予灌服四君子汤,模型组和正常组给予灌服等量的双蒸水,灌服4周后进行取材。麻醉动物,取下左侧腮腺,缝合伤口,等大鼠稍清醒时用10%冰乙酸溶液20μL刺激大鼠舌尖,1次/min,共刺激30min;处死,取下右侧腮腺。检测腮腺淀粉酶活性、电镜下观察腮腺内酶原颗粒数量、腮腺组织病理。结果模型组大鼠酸刺激后腮腺内残余的淀粉酶活性和酶原颗粒高于正常组大鼠,经四君子汤治疗后脾虚大鼠的唾液淀粉酶的分泌基本恢复正常,各组腮腺组织没有明显病理改变。结论四君子汤对利血平所致脾虚大鼠唾液淀粉酶分泌障碍有明显治疗作用,机制与腮腺组织病理学无关。
Objective To observe the effect of Sijunzi Decoction on secretion of salivary amylase in splenasthenic rats.Methods Male SD rats were divided into blank control group and reserpine model group.Rats in reserpine model group received subcutaneous injection of reserpine,while the rats in the control group received the same volume of saline for 8 days.After modeling,some rats in reserpine model group were given Sijunzi Decoction orally,and the other rats were fed the same amount of distilled water for 4 weeks.Rats in the blank control group were also given distilled water for 4 weeks.To study the reflex secretion,the animal was anaesthetized and the left parotid was removed.When the animals were just awake,20 μL of 10 % glacial acetic acid was applied on the tip of the tongue once every minute for 30 minutes,and then we executed the rats and removed the right parotid glands.The amylase activity was assayed,and the number of zymogen granules and pathological changes of glandular tissue were examined under electron microscope.Results After stimulated by glacial acetic acid,the amylase activity and the number of zymogen granules in the remained parotid salivary gland of model rats were increased as compared with the blank control group.The secretion of salivary amylase recovered to normal after treatment with Sijunzi Decoction.Parotid gland in each group showed no significant pathological changes.Conclution Sijunzi Decoction has obvious effect on deficiency of secretion of salivary amylase in in splenasthenic rat induced by reserpine.
