摘要
目的探讨青岛市汉坦病毒主要流行株的基因型。方法在青岛5市采集鼠肺标本,应用逆转录聚合酶链反应(RT-PCR)扩增技术及核苷酸序列测定方法对免疫荧光抗原阳性的HV M部分片段扩增并测序,与山东省其他地区代表株及其他部分标准株进行系统发生树及同源性分析。结果分离的5株HV有3株为SEO型,2株为HTN型。从SEO型病毒间的异同来看,青岛市汉坦病毒分离株和临近地区汉坦病毒分离株相近。结论青岛市HV是SEO和HTN混合型疫区;SEO型基因有较高的稳定性。
Objective To analyze the dominant genotypes of hantavirus in Qingdao.Methods Mice lung samples were collected from 5 regions in Qingdao.The RNA from lung samples positive for HV antigens according to immunofluorescence assay was amplified by RT-PCR with hantavirus-specific primers and the amplification products were sequenced.Phylogenetic analysis was used to compare the results with representative strains from other regions in Shandong and other standard strains.Results Five strains were positive for hantavirus.Homology analysis of nucleotide sequences showed that 2 strains were HTN HV and that the other 3 were SEO HV.The nucleotide sequences of the Qingdao SEO HV were homologous to adjacent regions.Conclusion SEO and HTN HV co-exist in Qingdao.SEO HV genes are highly conserved.
