摘要
目的:考察新疆紫花苜蓿总黄酮提取物的方法并进行黄酮含量测定。方法:用超声辅助的稀乙醇制备总黄酮提取物;以芦丁为对照品,用分光光度计在258.5nm波长处测定吸光度,计算提取物中黄酮含量。结果:超声辅助的60%乙醇提取苜蓿总黄酮的方法简单,具有黄酮提取率高、制备成本低、无污染等优点,适合于工业化生产;含量测定方法操作简便,重现性好,r=0.9998,平均加样回收率99.88%,RSD为1.35%;总黄酮平均含量为9.13%。结论:该实验方法可用于紫花苜蓿总黄酮提取物的制备和质量控制。
Objective:To envestigate the extraction and content determination methods of flavonoids of Medicago sativa L(alfalfa) plant originated from Xinjiang.Methods: The untrasonic ethanol extraction method was used for preparation of total flavonoid;the ultraviolet absorbances of the samples were measured with spectrophotometer at a wavelength of 258.5 nm with Rutin as a standard;and then the total flavonoid content was calculated.Results: The ultrasonic extraction with 60% of ethanol has some advanteges such as higher extraxtion rate,simple procedure and lower economic cost and contamination for total flavonoid preparation from alfalfa.Furthermore,this content determination method has a good linear relationship,r=0.9998,the average recovery rate 99.83%,RSD%=1.23%(n=9),the content of flavonoid in alfalfa extract was 9.13%.Conclusion: This method is suitable for the flavonoid extract and quality control of alfalfa plant and its preparation.
