摘要
研究了猪胰脂肪酶催化缩水甘油丁酸酯的对映选择性水解反应,考察了各种因素对酶促水解拆分过程的影响。结果表明,水解产物丁酸的大量生成会抑制脂肪酶的水解活性,至pH<5时酶促水解反应几乎停止。采用高浓度的磷酸盐缓冲液可有效地稳定系统的酸碱度,使酶促反应顺利进行。在缓冲液pH7.5,转速150r/min,反应温度30℃,每克底物酶用量25~35mg时,外消旋缩水甘油丁酸酯酶促水解转化率达60%,所得(R)-酯的对映体过量值(ee)为84.7%。
Optical resolution of racemic glycidyl butyrate has been efficiently achieved by lipase catalyzed enantioselective hydrolysis.The reaction conditions have been optimized.The optimum temperature for the enzyme reaction is around 30℃.The optimum initial pH of buffer is 7.5.However,too high concentration of butyric acid produced from the reaction would decrease the pH of the system and inhibit the hydrolytic activity of lipase.This problem was solved by using high concentration of phosphate as buffer system.The optical purity of the finally recovered ( R ) glycidyl butyrate was about 85% ee.
