放线菌327#的发酵培养基筛选及培养条件优化 被引量:6

摘要 为使放线菌327#最大限度地生产拮抗物质,在摇瓶试验中采用单因素和正交试验对其发酵培养基进行优化。同时考察了培养温度、初始pH值、发酵液装量对放线菌327#生产拮抗物质的影响。得到了最佳培养基配方为:大豆粉0.5%,硫酸铵1.5%,葡萄糖2.5%,氯化钠0.6%,碳酸钙0.6%。最佳发酵条件为培养温度30℃,初始pH值7.0,发酵液装量为250mL摇瓶装15mL。培养时间3d时,发酵液中拮抗物质达到最高水平,对青霉的抑菌圈直径达(22.4±0.4)mm。 The culture medium and fermentation conditions were investig ated for improving the production of antagonistic substances by Actinomycetes 327# in shaking flasks. By single factor and orthogonal experiment, the optimal fermentation medium of 327# were obtained as follows: soybean flour 0.5%, (NFL)2SO4 1.5%, glucose 2.5%, NaC1 0.6%, CaCO3 0.6%. The optimal fermentation conditions were as follows: culture temperature 30~C, initial pH 7.0, culture volume 15 ml in 250 mL shaking flask. Under these conditions, the inhibition zone diameter of 327# fermentation filtrate reached 22.4±0.4 mm on the test plates of PeniciUium after 3 d incubation.
