摘要 以米糠为原料,利用淀粉酶、纤维素酶和植酸酶3种非蛋白酶分步提取米糠蛋白,确定了最佳提取工艺条件。通过单因素试验和正交试验确定淀粉酶最佳提取条件为:淀粉酶添加量为20U/g,液固比8∶1,温度62℃,pH值6.2,提取时间2.5h,在此条件下,米糠蛋白质的提取率为56.16%。在利用淀粉酶水解米糠中的淀粉后,添加纤维素酶和植酸酶进一步水解米糠中的纤维素和植酸,以提高米糠蛋白的提取率,其中纤维素酶和植酸酶添加量分别为300,50U/g,在温度55℃,pH值为5.5下继续提取时间为2.5h。经3种非蛋白酶分步提取后,米糠蛋白的提取率达77.41%,蛋白的纯度达68.83%。 The rice bran is used as material for extracting rice bran protein with amylase,cellulase and phytase.During the process of extracting rice bran protein,the protein extracting ratio is as the index to confirm the effect on rice bran protein.As certaining the optimum decompose condition via single factor experiment and orthogonal experiment,the ratio of liquid to solid is 8:1, extracting time is 2.5 h,temperature is 62℃,pH is 6.2,the protease dosage is 20 U/g,the extraction rates of protein may reach 56.16%in this conditions.The rice bran first using the amylase at the most superior condition,then continues to use the cellulase and the phytase extracting rice bran protein when the enzyme recruitment respectively is 300,50 U/g,the temperature is 55℃,pH is 5.5,continues to extract 2.5 h.Using these three kind of non-proteinases,the rice bran protein's extraction rate may reach 77.41%,the protein purity may reach 68.83%.
