L-plastin启动子单核苷酸多态性与前列腺癌遗传易感性的相关性研究 被引量:2

Studies on the relationship between single nucleotide polymorphism of L-plastin gene promoter and susceptibility to prostate cancer

摘要 目的研究L-plastin启动子单核苷酸多态性(SNP)与前列腺癌的关系。方法采用Tm-shifting(等位基因特异性扩增结合融解温度曲线分析法)分析方法,对82例前列腺癌人群和94例对照人群的L-plastin启动子SNP位点的基因型进行检测。结果 L-plastin启动子TT、CT、CC基因型及等位基因T、C在汉族前列腺癌患者及对照者中的分布频率差异无显著性(P>0.05);在前列腺癌组内不同病理分级中,中、高分化组和低分化组中TT、CT、CC基因型分布频率差异无显著性(P>0.05);在前列腺癌组内不同临床分期中,与TT纯合子相比,CC纯合子前列腺癌Ⅳ期的风险增加至5.0倍(P<0.05)。结论 L-plastin启动子单核苷酸多态性在中国汉族人群中与前列腺癌无相关,但该启动子多态性可能在预测前列腺癌进展中有一定作用。 Objective To investigate the relationship between the single nucleotide polymorphism of L-plastin gene promoter and susceptibility to prostate cancer. Methods Eighty two PCa patients and ninety four age-matched controls were recruited in the study. The genotypes of single nucleotide polymorphism of L-plastin gene promoter were determined by real-time PCR based Tm-shifting method. Results There was no significant difference in the distribution of genotype and allele between PCa patients and normal controls (P〉0.05). In different histological grade of PCa group, the distribution of genotype had no significant difference between the intermediate-well differentiated subgroup and poorly differentiated subgroup. In different clinical stage of PCa group, there was 5.0-fold increased risk for the CC genotype in the stage IV PCa patients compared with the TT genotype in Chinese population (P〈0.05). Conclusion The results indicated that there was no significant relationship between the single nucleotide polymorphism of L-plastin gene promoter and susceptibility to PCa in Chinese Han population. However, this SNP genotype of L- plastin gene promoter might be associated with the progression of prostate cancer.
