呋塞米对三聚氰胺与三聚氰酸致大鼠肾损伤的干预研究

摘要 目的:研究呋塞米对三聚氰胺与三聚氰酸致大鼠肾脏损伤的干预作用。方法:选用40只成年雄性SD大鼠,体重200~220g,随机分为4组,每组10只,分别为A组(空白对照组)、B组(呋塞米20mg/kg)、C组(三聚氰胺与三聚氰酸各100mg/kg+呋塞米20mg/kg)、D组(三聚氰胺与三聚氰酸各100mg/kg)。每天每只大鼠灌胃一次,于4d后观察大鼠肾脏病理,肾结晶沉积情况、大鼠血和尿多项生化指标的变化。结果:在D组,病理切片发现许多黄色圆形晶体,这些晶体集聚堵塞于肾小管中,造成肾小管明显扩张、上皮细胞肿胀等病理改变。与D组相比,C组结晶评分下降了56.76%,尿量、尿酸排泄量、尿钠、尿钾排泄量显著增加(P<0.05);血清尿素氮、肌酐、血钠、血钾显著减低(P<0.05)。D组和其余各组相比,血清尿素氮、肌酐、血钠、血钾显著升高(P<0.05),尿量、尿酸排泄量、尿钠、尿钾排泄量显著减少(P<0.05)。结论:三聚氰胺与三聚氰酸联合可致大鼠肾损伤,主要是通过结晶沉淀造成急性梗阻所致,呋塞米对三聚氰胺与三聚氰酸所致肾损伤起一定保护作用。
