维生素D_3对IL-4和TNF-α诱导的气道平滑肌细胞TARC表达的影响 被引量:2

摘要 目的:探讨维生素D3(vitaminD3,VD3)对白介素4(IL-4)和肿瘤坏死因子α(TNF-α)诱导的气道平滑肌细胞(ASMCs)胸腺和活化调节趋化因子(thymus and activated regulation chemokine,TARC)表达的影响。方法:体外培养ASMCs,给予IL-4和TNF-α诱导后分为5组:空白对照组(A组)、IL-4+TNF-α诱导组(B组)、地塞米松干预组(C组)、VD3干预组(D组)、地塞米松+VD3干预组(E组),采用酶联免疫吸附法(ELISA)测定细胞上清液中TARC和嗜酸性粒细胞趋化因子(eotaxin)的表达,逆转录聚合酶链反应(RT-PCR)法检测TARC mRNA的表达。结果:(1)ASMCs培养上清液中TARC和eotaxin的浓度,B组较A组表达明显增加,C、D、E组较B组表达降低(均P<0.05)。(2)TARC mRNA的表达,B组较A组表达明显增加,C、D、E组较B组降低,较A组升高(均P<0.05)。结论:VD3抑制了ASMCs内TARC的表达,降低了细胞因子eotaxin的表达水平。
