风湿宁对佐剂性关节炎大鼠关节破坏的保护作用

Effect of Feng-Shi-Ning on Regulating the Expression of uPA and MMP-3 in Adjunvant Arthritis Rats

摘要 目的:探讨中药复方风湿宁(FSN)对实验性类风湿性关节炎(RA)的疗效及作用机理。方法:以佐剂性关节炎(adjunvant arthritis,AA)大鼠为实验对象,分别予以FSN与雷公藤多甙(TWP)进行治疗,对照观察大鼠关节内病理组织学变化情况。采用免疫组织化学检测各组AA大鼠病变关节组织中uPA、MMP-3的表达。结果:FSN可显著抑制AA大鼠病变关节滑膜炎症与增生,防止关节软骨及骨质的破坏;FSN能明显降低uPA、MMP-3在AA大鼠软骨、骨组织及滑膜组织的表达(P<0.05或P<0.01)结论:FSN具有防止RA关节破坏的作用,其作用与阻断蛋白酶系统对软骨、骨基质成分的降解有关。 Objective:To observe the therapeutic effect of Feng-Shi-Ning(FSN) against adjuvant-induced arthritis in rats,as well as the mechanism.Methods:Take the adjunvant arthritis(AA) rats as animal model for RA,which were treated with FSN and tripterygium wilfordii polyglycosidum(TWP) respectively.And the expression of uPA and MMP-3 were observed by immunohistochemical examination.Results:FSN could prevent the joint cartilage and bone from destruction.The expression of uPA and MMP-3 were observed on synovium and chondrocyte.FSN could decrease the leveles of uPA and MMP-3(P0.01).Conclusions:The data showed that FSN has inhibitory effects on symptom and condition of experimental RA,the anti-inflammatory effect is related to its adjustment effect on the expression of MMP-3 and uPA.
