二巯丙磺钠对溴苯腈急性中毒心肌能量代谢影响

Effects of Na-DMPS on energy metabolism of myocardium in mouse with acute bromoxynil toxicity

摘要 目的探讨溴苯腈急性中毒对小鼠心肌能量代谢的影响及二巯丙磺钠(Na-DMPS)的保护作用。方法 ICR小鼠78只,随机分成5组,对照组、溴苯腈高、低剂量组和Na-DMPS干预组。检测染毒后0.5,3 h心肌腺苷酸含量。结果溴苯腈高剂量组染毒后0.5,3 h,三磷酸腺苷(ATP)含量分别为(39.62±8.39),(41.19±6.56)mg/g,明显低于对照组(49.96±6.87)mg/g(P<0.05);二磷酸腺苷(ADP)含量在染毒后0.5,3 h高于对照组(11.01±0.93)mg/g(P<0.05);磷酸腺苷(AMP)于染毒后3,6 h高于对照组(1.61±0.20)mg/g;Na-MDPS干预后ATP分别为(51.10±7.10),(58.34±12.34),(52.92±14.15)mg/g,明显高于溴苯腈高剂量组(P<0.05);ADP、AMP含量低于溴苯腈高剂量组(P<0.01)。结论溴苯腈急性中毒可影响小鼠心肌能量代谢,Na-DMPS干预可改善心肌能量代谢。 Objective To explore the effects of sodium dimercaptopropane sulfonate(Na-DMPS) on energy metabolism of myocardium with acute bromoxynil toxicity.Methods Seventy-eight ICR mice were randomly divided into normal control group(n=6),bromoxynil poisoning group(n=36),and Na-DMPS protection group(n=36).The mice in bromoxynil poisoning group and Na-DMPS protection group were sub-dvvided randomly in to high and low dose group.The contents of adenosine triphosphate(ATP),adenosine diphosphate(ADP),and adenosine monophosphate(AMP) in myocardium were detected at 0.5,3,and 6 hours after the treatment.Results The ATP contents were 39.62±8.39,41.19±6.56 mg/g at 0.5,3 hours,respectively,in mice of high dose bromoxynil exposure group and significantly lower than those of control group(P0.05).The contents of ADP at 0.5,3 hours after high-dose bromoxynil exposure were significantly higher than that of control group(14.73±3.20 and 17.28±2.36 vs 11.01±0.93 mg/g,P0.05).The contents of AMP in mice with high dose bromoxynil exposure at 3,6 hours after the treatment were higher than that of control.The ATP contents in mice with Na-DMPS exposure were 51.01±7.10,58.34±12.34,and 52.92±14.15 mg/g at 0.5,3,and 6 hours after the treatment,respectively,and significantly higher than those of mice in high-dose bromoxynil group(P0.05).But the contents of ADP and AMP in mice of Na-DMPS group were lower than that of high-dose bromoxynil group(P0.01).Conclusion Acute bromoxynil intoxication affects myocadium energy metabolism and Na-DMPS intervention improves myocardium energy metabolism in mice with the intoxication.
