摘要 以电化学法和光度法对铬黑T(Eriochrome black T,EBT)与牛血清白蛋白(Bovine serum albumin,BSA)的相互作用进行了研究。研究表明:在pH5.29的(Britton-Robinson,B-R)缓冲液中,EBT有两个氧化峰和两个还原峰,加入BSA后EBT峰电位不变,峰电流下降,据此建立了一种测定BSA的电分析方法。在优化条件下,峰电流下降值与所加的BSA量在2.0×10-6~1.0×10-5mol/L、2.0×10-6~6.0×10-7mol/L和6.0×10-7~5.0×10-8mol/L范围内呈线性响应,检测限为1.0×10-9mol/L。通过测定BSA引入前后的一些电化学参数,推测EBT与BSA相互作用生成了一种非电活性的超分子化合物,探讨了BSA与EBT的结合反应机理。 The interaction of erichrome black T(EBT) with bovine serum albumin(BSA) was studied by electrochemical method and spectrophotometry.EBT showed two oxidation peaks and two reduction peaks.Upon addition of BSA to the above solution,the peak current of EBT decreased without changes in the peak potential.An electrochemical method for quantitative analysis of BSA was developed based on the interaction of EBT with BSA.Under the optimized condition,the decrease of the peak currents of EBT versus BSA concentration gave rise to three linear regression equations in the BSA concentration range of 2.0×10-6~1.0×10-5 mol/L,2.0×10-6~6.0×10-7 mol/L,and 6.0×10-7~5.0×10-8 mol/L,respectively,with the detection limit equating to 1.0×10-9 mol/L.The electrochemical parameters in the absence and presence of BSA were estimated.An electrochemical inactive supermolecular compound BSA-EBT was formed.Furthermore,the binding mechanism was also discussed.
