摘要
目的调查多药耐药鲍氏不动杆菌(MDR-ABA)转座子、整合子等可移动遗传元件的存在情况。方法收集宁波市李惠利医院2009年6-9月住院患者中分离的MDR-ABA共20株,采用聚合酶联反应(PCR)及序列分析的方法分析10种可移动遗传元件:tnpU、tnp513、IS26、ISEcp1、ISaba1、ISaba4、ISaba9、intⅠ1、intⅠ2、intⅠ3。结果 20株MDR-ABA共检出5种基因:tnpU、tnp513、IS26、ISaba1、intⅠ1,其余5种基因未检出;各种基因阳性率以IS26和ISaba1最高16株(80%),intⅠ1次之13株(65%),tnp513为12株(60%),tnpU9株(45%);9株MDR-ABA同时检出5种基因,1株MDR-ABA同时检出4种基因,2株MDR-ABA同时检出3种基因,5株MDR-ABA同时检出2种基因,1株MDR-ABA仅检出1种基因,另有2株MDR-ABA未检出上述10种基因。结论同时检测MDR-ABA10种可移动遗传元件是国内首次报道,可移动遗传元件介导各种耐药基因使受体菌表现为多药耐药。
OBJECTIVE To investigate the distribution of mobile genetic elements:transposons and integrons in multidrug-resistant Acinetobacter baumannii(MDR-ABA).METHODS From Jun 2009 to Sep 2009,20 strains of MDR-ABA were collected from Ningbo Li Hui-li Hospital,then,10 kinds of mobile genetic elements(tnpU,tnp513,IS26,ISEcp1,ISaba1,ISaba4,ISaba9,intⅠ1,intⅠ2 and intⅠ3)were analyzed by PCR and verified by DNA sequencing.RESULTS In 20 strains of MDR-ABA,16 strains(80%)were detected to carry IS26 and ISaba1,13 strains(65%),12 strains(60%)and 9 strains(45%)were detected to carry intⅠ1,tnp513 and tnpU,respectively.However,the other 5 kinds of genes were not detected out.In addition,9 strains were detected out to carry 5 kinds of genes,1 strain was detected to carry 4 kinds of genes,2 strains were detected to carry 3 kinds of genes,5 strains were detected to carry 2 kinds of genes,1 strain was detected to carry 1 kind of gene,2 strains couldn't be detected to carry any kind of genes.CONCLUSION It's the first report to detect 10 kinds of mobile genetic elements at the same time in China.And acquired resistant genes induced by mobile genetic elements cause the recipient strains to express multi-drug resistance.
