摘要
目的:通过慢性长波紫外线(UVA)照射小鼠皮肤形成光老化损伤,探讨强脉冲光(IPL)改善皮肤光老化的可能作用机制。方法:将实验所用BALB/c小鼠按不同处理条件分为6组:空白组1、UVA组、8-甲氧补骨脂素(8-MOP)+UVA组、UVA+IPL组、8-MOP+UVA+IPL组和空白组2。UVA或8-MOP+UVA连续照射12周,继之IPL照光2次,间隔2周。取小鼠皮肤组织行组织病理检查及透射电镜检查。结果:与空白组比较,UVA组小鼠皮肤真皮层变厚,可见胶原组织变性。8-MOP+UVA组小鼠皮肤表皮层亦见增厚,真皮层厚度的增加和异常胶原纤维的堆积较UVA组更显著,电镜下可见线粒体空泡化变性,胶原纤维溶解。IPL处理后,小鼠皮肤表皮层未表现明显增厚,真皮层扭曲变性的胶原纤维结构得到改善,胶原纤维排列重新趋于整齐紧密,电镜下组织受损明显得到改善。结论:8-MOP可以加速促进光老化进程,慢性UVA照射+8-MOP可成功快速诱导小鼠皮肤的光老化损伤。IPL治疗能够有效改善小鼠皮肤光老化。
Objective: To investigate the improving effects of intense pulse light (IPL) on the skin of photoageing model mice induced by long-term ultraviolet A or 8-MOP+UVA irradiation. Methods: The BALB/e mice were randomly divided into 6 groups with different experimental intervals: control group 1, UVA group, 8-MOP+UVA group, UVA+IPL group, 8-MOP+UVA +IPL group and control group 2. After 8-MOP+UVA and/or IPL treatment for terminals, the irradiated skin specimens were taken for histopathology ex amination by light microscopy and electron microscopy. Results: Compared with the control group, the epidermis showed a little thick- er and the collagen tissue in the dermis became degenerated in UVA group. These changes became more remarkable in 8-MOP+UVA group. Under the intervention of IPL, the photoaging changes of skin tissue could be improved obviously. Conclusions: The murine photodamage mouse skin can be established better and effectively by means of 8-MOP+UVA. IPL has the protective potency on the skin of photoaging mice.
