黄芪总苷诱导人白血病NB4细胞凋亡的研究

Study on the apoptosis of NB4 cells induced by total astragalosides

目的研究黄芪总苷(TA)对体外培养的人早幼粒细胞白血病细胞株NB4凋亡的影响。方法将不同浓度的TA(200、400、600、800μg/ml)作用于NB4细胞,继续培养48h,采用磷脂结合蛋白V(AnnexinV)+碘化丙啶(PI)双染色法在流式细胞仪上检测细胞凋亡率。结果 TA作用于NB4细胞48h后,各浓度组的凋亡率与对照组相比差异均有统计学意义(P<0.05)。在200、400、600μg/ml组凋亡率呈浓度依赖性增加,各组之间差异有统计学意义(P<0.05)。在800μg/ml组凋亡率增加不明显,与600μg/ml组比较差异无统计学意义(P>0.05),但出现了大量的坏死细胞。结论一定剂量范围内的TA可以诱导NB4细胞凋亡,具有体外抗白血病效应。

Objective To study the effect of total astragalosides（ TA） on the apoptosis of NB4 cells. Methods The NB4 cells were treated with TA of different concentration （ 200,400,600,800 μg/ml） for 48 h in culture. The apoptosis of NB4 cells marked with Annexin V and PI double staining was detected by flow cytometry. Results After incubation for 48 h,there was significant difference in apoptosis between TA group and blank control（ P 0. 05） . Besides,TA induced apoptosis of NB4 cells in a dose-dependent manner in groups of 200 μg/ml,400 μg/ml,600 μg/ml（ P 0. 05） . But there was no significant difference in apoptotic rates between the group of 800 μg/ml and 600 μg/ml（ P 0. 05） . In the group of 800 μg/ml,the necrotic cells elevated highly. Conclusion In some dose limit,TA can induce apoptosis of NB4 cells. Consequently,TA has an antileukemie effect in vitro.
