一氧化氮对大鼠背根神经节瘦素长型受体表达的影响

摘要 目的研究一氧化氮(NO)对大鼠背根神经节(DRG)神经元中瘦素长型受体(OB-Rb)mRNA和蛋白表达的影响。方法用一氧化氮供体硝普钠(SNP;0、0.01、0.1和0.5 mmol/L)处理体外培养大鼠DRG神经元;用RT-PCR和免疫印迹技术检测神经元中瘦素长型受体mRNA和蛋白的表达。结果0.1~0.5 mmol/L的SNP能促进瘦素长型受体mRNA和蛋白表达。结论NO对DRG的功能调节可能与瘦素相关。 Objective To investigate the effect of nitric oxide(NO) on the expression of long type leptin receptor(OB-Rb) in dorsal root ganglion(DRG) neurons.Methods Sodium nitroprusside(SNP;0,0.01,0.1 and 0.5 mmol/L) were used to treat the DRG neurons cultured in vitro.RT-PCR and Western blot were used to detect the expression of long types of leptin receptor(OB-Rb) in DRG neurons.Results 0.1~0.5 mmol/L SNP can increase the expression of OB-Rb mRNA and protein.Conclusion The modulation of NO on DRG neurons may be related with the function of leptin.
