一氧化氮抑制胃癌细胞系SGC-7901增殖及C-FOS和C-JUN表达

摘要 目的探讨一氧化氮(NO)对胃癌细胞系SGC-7901增殖的影响及其机制。方法MTT法测定细胞的生长,RT-PCR和W estern印迹法检测C-FOS和C-JUN的mRNA和蛋白表达。结果硝普钠(SNP)抑制SGC-7901细胞的生长(P<0.05),并呈剂量和时间性降低C-FOS和C-JUNmRNA及蛋白表达。结论NO可能通过下调C-FOS和C-JUN的表达而抑制胃癌细胞的增殖。 Objective To investigate the effect of nitric oxide(NO) on cell proliferation of gastric cancer cell line SGC-7901,and to investigate the relevant mechanisms.Methods MTT assay was used to detect the inhibitory effect of NO on cell growth.RT-PCR was used to detect the mRNA expression of C-FOS and C-JUN.Western blot was applied to study the protein expression of C-FOS and C-JUN.Results Sodium nitroprusside(SNP) inhibited the proliferation of SGC-7901 cells(P0.05).SNP dose-and time-dependently decreased the expression of C-FOS,C-JUN mRNA and protein.Conclusion NO inhibits the proliferation of SGC-7901 cells,which is probably down-regulated C-FOS and C-JUN expression through NO.
