纳洛酮对少突胶质细胞的保护作用

摘要 目的研究纳洛酮(naloxone)对脂多糖(lipopolysacchar,LPS)激活的小胶质细胞诱导少突胶质细胞损伤的影响。方法共培养大鼠小胶质细胞和少突胶质细胞,用LPS(1μg.mL-1)刺激小胶质细胞,随后用0.1μM纳洛酮进行干预,应用免疫组化方法观察计数少突胶质细胞。结果仅用0.1μM纳洛酮处理细胞,细胞活性基本保持不变,而当小胶质细胞被LPS激活后,与之共培养的大量的少突胶质细胞死亡(P<0.01);小胶质细胞被LPS激活,给予0.1μM纳洛酮处理后,少突胶质细胞的活细胞数显著升高(P<0.05)。结论纳洛酮能够抑制LPS激活的小胶质细胞引起的少突胶质细胞的损伤。 Objective To explore the protective effect of naloxone on oligodendrocytes injury caused by LPS activated microglia cells.Methods Microglia cells cocultured with oligodendrocytes were treated with LPS(1μg·mL-1),then treated with 0.1μM naloxone for 18 h.The protective effect of naloxone was measured by IHC staining.Results After microglia cells were activated by LPS,the injury of cocultured oligodendrocytes could be obviously shown.However,treatment of LPS activated microglia cells with 0.1μM naloxone significantly prevented against the oligodendrocytes injury caused by microglia activation.Conclusion Although naloxone can protect oligodendrocytes injury from activated microglia cells,it would be a good candidate for treating some neurodegenerative diseases.
