摘要
目的观察Intermedin(IMD)在两肾一夹(2-kidney1-clip,2K1C)高血压大鼠肾脏和腹主动脉中的变化及其意义。方法采用2K1C复制肾性高血压大鼠模型,随机分为正常组、模型组。分别测定平均颈动脉压(mCAP)、左心室舒张末期压(LVEDP)、左心室压力变化最大速率(LV±dp/dtmax),左心重与体重比值(LVM/BW)及右肾重与左肾重比值(RK/LK)。免疫组化法检测IMD的水平。结果模型组大鼠mCAP和LVEDP比正常组升高48.23%、340%(P均<0.01),LV+dp/dtmax和LV-dp/dtmax降低51.26%和24.65%(P均<0.01),LVM/BW以及RK/LK升高36.77%(P<0.05)、26.35%(P<0.01);与正常组比较模型组肾脏和腹主动脉IMD的含量分别增加了26.15倍和4.80倍(均P<0.01)。结论肾性高血压大鼠引起肾脏和腹主动脉IMD蛋白表达增加,其变化可能参与了肾性高血压的发病过程。
Objective To observe the changes and significance of Intermedin(IMD) in two-kidney-one-clip(2-kidney1-clip,2K1C) hypertensive rats' kidney and abdominal aorta.Methods The replications of 2K1C renal hypertensive rats were randomly divided into normal group and model group.Mean carotid arterial pressure(mCAP),left ventricular end-diastolic pressure(LVEDP),maximum rate of left ventricular pressure(LV±dp/dtmax),left ventricular weight to body weight ratio(LVM/BW),right kidney weight and left kidney weight ratio(RK/LK)were measured.Immunohistochemical detection of IMD's level were performed.Results mCAP and LVEDP increased 48.23% and 340% in the model group than those in the normal group(all P0.01).Compared with the normal group,LV+dp/dtmax and LV-dp/dtmax reduced 51.26% and 24.65% in the model group(all P 0.01),LVM/BW and RK/LK increased 36.77% in model group(P0.05) and 26.35%(P0.01).Compared with normal group,the content of IMD of kidney and abdominal aorta of model group increased by 26.15 times and 4.80 times respectively(all P0.01).Conclusion The renal hypertension increased the IMD's protein expression in kidney and abdominal aorta and the changes might be involved in the pathogenesis of renal hypertension.
