副溶血弧菌中具有Mg^(2+)转运功能的MgtE基因的分子克隆与功能鉴定

摘要 MgtE基因家族是最初在细菌中发现的一类具有Mg2+转运能力的基因家族。基因组数据库搜索显示副溶血弧菌(Vibrio parahaaemolyticus RIMD 2210633,VP)中存在一个MgtE基因家族成员。克隆了该基因并将之命名为VP-MgtE,并利用沙门氏杆菌突变株MM281作为宿主菌进行了功能互补实验,同时运用定点突变的方法研究了VP-MgtE中的保守氨基酸对其功能的影响。 MgtE gene family,which has the ability of Mg2+ transport,was first found in bacteria.Genomic sequence data indicated the presence of MgtE homolog in the gram-negative bacterial Vibrio parahaemolyticus.We cloned it and named it as VP-MgtE,and the functionally complement experiments of VP-MgtE were conducted to a salmonella mutant MM281 which was as host bacteria.Meanwhile,the impact of conservative amino acid in VP-MgtE on the functions of VP-MgtE was studied by site-directed mutagenesis method.
