摘要 Tristetraprolin(TTP)是一种RNA结合蛋白,能与mRNA 3′非翻译区富含AU区域结合,介导mRNA的降解.诸多研究显示,TTP对急性肺损伤中重要炎症介质的表达发挥了关键的调节作用.许多因素可以调控TTP的磷酸化状态以及亚细胞定位,其中p38 MAPK激活蛋白激酶2可以使TTP磷酸化,而蛋白磷酸酶2A可使TTP去磷酸化,通过调节TTP的磷酸化状态以及亚细胞定位影响其降解mRNA的功能和相应炎症介质的表达. Tristetraprolin(TTP) is an RNA -binding protein that binds to an AU -rich element in the 3'- untranslated region of its target mRNAs and accelerates the degradation of mRNAs. Many studies showed that TTP played a critical role in the regulation of inflammatory mediators expression in acute lung injury. A series of factors could regulate the phosphorylation state and subcellular localisation of TTP,of whom p38 MAPK activated protein kinase- 2 phosphorylates TFP, while Protein Phosphatase 2A dephosphorylates TTP, then they influenced the function of TTP- mediated mRNA decay and the inflammatory mediators expression.
