摘要
以桂皮紫萁的穗状孢子为外植体进行离体快繁研究。结果表明,破碎孢子囊后的孢子接种在1/6MS+蛋白胨0.5g/L+蔗糖20g/L+琼脂8.6g/L中,在变温变光下培养更易萌发。1/2MS培养基中添加8g/L桂皮紫萁叶片提取液以及(KT5.0mg/L和NAA0.5mg/L、NAA1.0mg/L)均能更好的促进原叶体的增殖。变温变光培养有利于瓶内孢子体的形成和生长,培养瓶外诱导更适合孢子体的转化。
This article was about the rapid reproduction of Osmurtda cinnamomea L. var. asiatica using the fringy sporophyll as explant in vitro. The results showed that the spores from broken sporangia inoculated in medium of 1/ 6MS with peptone 0.5g/L + sucrose 20g/L + agar 8.6g/L were easy to germinate quickly under the c, ondition of changeable temperature and light. 1/2MS culture medium added with 8g/L liquid extracted from leaves cff Osmunda cinnamomea L. var. asiatica or KT 5.0mg/L, NAA 0.5mg/L and NAA 1.0mg/L,both of which presented good performance in promoting proliferation of prothallium. The changeable condition of different temperature and light bene- fited the growth of sporophyte in vitro. Culture in persi dishes rather in bottles would be more suitable for prothallus transforming into sporophyte.
