甘蔗耐盐愈伤组织的离体筛选及鉴定 被引量:2

摘要 以甘蔗心叶为外植体,MS+2,4-D2mg/L为基本培养基,在25±2℃、光照强度24~30μmol/m2.s、每天光照10h条件下进行耐盐愈伤组织的离体筛选和鉴定。结果表明,甘蔗愈伤组织的半致死和致死盐浓度分别为0.5%、1.5%NaCl,其愈伤组织诱导率分别为57.5%、0,极显著低于对照;在最高筛选盐浓度(1.0%NaCl)胁迫下,耐盐愈伤组织的相对生长量(2.363%)极显著低于对照(3.162%);耐盐植株的脯氨酸含量(527.19ug/g)极显著高于对照(185.06ug/g),相对电导率(32.3)极显著低于对照(49.0)。 The central leaves of Saccharum officenarum L. as explants were 2,4-D 2mg/L. In vitro selection of salt tolerant callus and identification of salt ditions of 25 ± 2 ,illumination intensity of 24 -30μmol/m^2. s and period cultivated on MS basic medium with tolerance were of lOh/d. The studied under the conresult showed that the semi-lethal and lethal salt concentration of sugar cane were 0.5% and 1.5%, respectively. The induction rates were 57.5% and 0. Under the 1.0% NaCl salt stress, the relative growth of callus (2. 363 ) was significantly lower than CK(3. 162),proline contents of salt-tolerant individual (527.19ug/g) were significantly higher than CK( 185.06 ug/g), and the relative conductivity of salt tolerant mutant (32.3) were significantly lower than CK(49.0).
