摘要
目的寻找导致MES(Matsumoto Eosinophilic Shinshu)大鼠嗜酸性粒细胞增多的致病基因和阐明发病机制。方法利用逆转录-聚合酶链反应,测序的方法,比较了MES大鼠和ACI大鼠位于19号染色体长臂末端的多个候选基因,并对其相应酶的活性进行了测定。结果在MES中,cyba基因转录mRNA在第4和第5外显子之间插入51个碱基,从而导致相应NADPH氧化酶的活性降低了49.86%。结论 cyba变异和NADPH氧化酶的活性降低可能与MES大鼠的嗜酸性粒细胞增多的发病机制有关。
Objective To find causal gene and elucidate the mechanism (Matsumoto Eosinophilic Shinshu ) rat. Methods Examine the positional of eosinophilia in MES candidate genes were examined using RT - PCR and sequence , and then the activity of corresponding enzyme was mensured. Results cyba gene was inserted 51 - bp between exon 4 and exon 5 in MES rat. NADPH oxidase activity decreased 50% in MES rat. Conclusion Mutation in cyba and decrease in NADPH oxidase activity might be involved in the mechanism of eosinophilia in MES rat.
