喉鳞状细胞癌中MMP1和TIMP3表达的相关性分析

摘要 目的探讨喉鳞状细胞癌(laryngeal squamous cell carcinoma,LSCC)中基质金属蛋白酶1(matrixmetalloproteinase 1,MMP1)和基质金属蛋白酶抑制因子3(tissue inhibitor of metalloproteinase 3,TIMP3)mRNA和蛋白的表达,探讨MMP1和TIMP3在LSCC发展和监测中的意义。方法分别应用逆转录-聚合酶链反应及免疫组织化学SP法,检测LSCC组织中MMP1和TIMP3 mRNA及蛋白表达情况。结果 MMP1 mRNA在LSCC组织中的表达0.66±0.26高于癌旁组织0.49±0.23(P<0.05),TIMP3 mRNA在LSCC组织中的表达0.34±0.21低于癌旁组织0.49±0.24(P<0.05);MMP1在LSCC组织和癌旁组织中的蛋白表达阳性率分别为82.4%和18.2%,两者比较差异有统计学意义(P<0.05);TIMP3在LSCC组织和癌旁组织中的蛋白表达阳性率分别为80.4%和100%,两者比较差异有统计学意义(P<0.05);MMP1和TIMP3 mRNA及蛋白表达与组织病理学分级、淋巴结转移相关(P<0.05),而与年龄无关(P>0.05);LSCC组织中MMP1和TIMP3 mRNA及蛋白表达未检测到相关性(P>0.05)。结论 MMP1和TIMP3表达与LSCC的发生发展、浸润及转移有关,检测LSCC组织中的MMP1和TIMP3的基因及蛋白表达情况,有助于判断LSCC的转移、TNM分期及预后情况。 Objective To investigate the mRNA and protein expression of matrix metalloproteinase 1 (MMPI) and tissue inhibitor of metalloproteinase 3 (TIMP3)in laryngeal squamous cell carcinoma (LSCC). Methods RT - PCR and immunohistochemistry SP method were used respectively to determine the mRNA and protein expression of MMP1 and TIMP3 in laryngeal squamous cell carcinoma. Results The mRNA expression of MMP1 in tumor tissues 0.66 ± 0.26 was higher than that in corresponding non - cancerous tissues 0.49± 0.23 ( P 〈 0.05 ) , mRNA expression of TIMP3 in tumor tissues 0.34 ±0.21 was lower than that in corresponding non - cancerous tissues 0.49 ±0.24 ( P 〈 0.05). The positive protein expression of MMP1 in the tumor tissues of LSCC (82.4%) was higher than that in corresponding noncancerous tissues (18.2%, P 〈 0, 05 ), the positive protein expression of TIMP3 in the tumor tissues of LSCC (80.4%) was lower than that in corresponding non - cancerous tissues ( 100%, P 〈 0.05). The mRNA and protein expression of MMP1 and TIMP3 was correlated with the development of laryngeal squamous cell carcinoma, grade and regional lymph node metastasis ( P 〈 0.05). Conclusion MMP1 and TIMP3 expression is associated with the development, infiltration and metastasis of laryngeal squamous cell carcinoma. Detection of the MMP1 and TIMP3 gene and protein expression in laryngeal cancer will help to determine metastasis, TNM stage and prognosis of laryngeal squamous cell carcinoma.
